I'd email instead of calling and would probably say "how many people will be there?"

How many people were invited? How many people will be present? How many people do you expect? Many options.

How many of you are there? The other question just doesn't seem right. Plus, I would never actually do this, so I wouldn't venture an "other" guess.

"How many of you are there?" but I would be more likely to say something like "And how large is the party?" or "How big is the group?"
